Reconstitution and concentration in the laboratory

The short answer

Reconstitution is bringing a lyophilised solid into solution with a solvent. Concentration is simply the mass of peptide divided by the volume of solvent added, and it is a laboratory calculation, nothing more.

What reconstitution is

Lyophilised peptide is supplied as a freeze-dried solid because it is far more stable that way. To work with it in solution, a solvent is added and the solid dissolves. That is reconstitution.

The usual solvent for research handling is bacteriostatic water: sterile water containing 0.9% benzyl alcohol as a preservative. The preservative is what allows a multi-use vial to hold up in a refrigerator for around 28 days instead of a few days.

Concentration is arithmetic

Concentration is mass divided by volume. Add 2ml of solvent to a 10mg vial and the solution is 5mg per ml. Add 1ml to the same vial and it is 10mg per ml. That is the whole calculation.

The choice of volume is a laboratory decision about the working range of the equipment and the precision required. A more dilute solution makes small volumes easier to measure accurately. A more concentrated one makes the vial last longer.

Technique that protects the material

Bring the vial and the solvent to room temperature first. Cold glass condenses atmospheric moisture into material that needs to stay dry.

Run the solvent slowly down the inner wall of the vial. Do not fire it directly into the lyophilised cake.

Do not shake. Agitation at the air-liquid interface denatures peptides. Roll the vial gently between the fingers and let it dissolve in its own time.

Aliquot immediately into single-use portions and freeze them. This is what protects the material from cumulative freeze-thaw damage.
